Investigating the Binding Mode of Ligands to α-Syn Proteins

SynuSight has resolved the high-resolution structures of a variety of compounds with α-Syn fibers. The results show that small molecule ligands exhibit three distinct conformational relationships when binding to fibers: diagonal geometry, horizontal geometry, and vertical geometry. Compared with conventional protein-ligand interactions, these binding patterns reveal a key feature - the π-π stacking between small molecule ligands is decisive for their specificity in binding amyloid fibrils.

A New Paradigm of Structure-Driven Drug Design

Building upon profound structural biology insights, SynuSight has pioneered a novel "Structure-Driven Drug Design" paradigm. By integrating AI algorithms with computer-aided drug design (CADD) technologies, we have achieved de novo development of a next-generation tracer featuring a novel molecular scaffold. This compound demonstrates both high specificity for α-Syn binding and exceptional blood-brain barrier (BBB) penetration with validated imaging performance in clinical studies.
